In the case where you are checking the probable deformation of concrete. For eg, if you remove the formwork for a structural slab before 28 days, you might need to verify the permanent deformation that occurs. As you know, E plays a big role in the determination of delta.

The modulus of elasticity of concrete at younger ages 28dias can be obtained by knowing the maturity curve that describes it.

Knowing the slope of the maturity of the concrete, it is possible to obtain the resistance of the same feature (fc) for times smaller or larger than 28días.

Then apply the same formula set forth in this site .. but now calculated using the fc curve maturation.

According to ASTM C469, why do we used the value of stress at 40% of ultimate load for calculating the modulus of elasticity?

Because as you know the material concrete has no linear stress strain diagram. Therefore I guess the ASTM has chosen the slope of that curve at 40% to be the best fitting slope since the slope is changing though all different strains and loads.
